It is the S-enantiomer of ketamine and acts as an NMDA receptor antagonist. Esketamine has been approved by the US FDA and regulatory bodies in multiple regions for the treatment of treatment-resistant depression (TRD) and major depressive disorder with acute suicidal ideation or behaviour.

- 已嘗試至少兩種不同抗抑鬱藥物（足夠劑量及療程），但療效不佳的難治型抑鬱症患者 已尝试至少两种不同抗抑郁药物（足够剂量及疗程），但疗效不佳的难治型抑郁症患者 Patients with treatment-resistant depression who have tried at least two different antidepressants at adequate doses without sufficient response
- 伴有急性自殺意念或行為的重度抑鬱症患者 伴有急性自杀意念或行为的重度抑郁症患者 Patients with major depressive disorder with acute suicidal ideation or behaviour
